NewLimit is composed of collaborative teams that each focus on a specific Technology or Product competency. Each of our goals is spearheaded by one of our teams, but every goal requires collaboration across all of our teams to achieve.

Three of our teams — Read, Write, and Predict — form our Discovery Engine. The Engine teams provide a set of technical competencies that enable reprogramming factor discovery across diverse product areas. Our Immunology team represents the first of our Product teams focused on developing a reprogramming medicine to address a particular indication. Over time, we will add additional Product teams to diversify our pipeline.

The roles and responsibilities of our current teams are outlined below.

Read

Descriptive Name: Single Cell Technology

Objective: Enable NewLimit to “read” the epigenetic state of target cell populations at large scale, low cost, and high quality

Competencies: Single cell and bulk functional genomics, sample preparation, molecular biology

Team Level Key Performance Indicators:

• Quality of single cell profiles captured in target cell types (e.g. number of unique seq reads, number of unique genes/peaks, fraction of peaks in functional regions)
• Cost per cell of single cell profiling
• Perturbation barcode capture rate

Write

Descriptive Name: Epigenetic Editing

Objective: Enable NewLimit to “write” epigenetic states in target cells by reprogramming using high-throughput assays

Competencies: Pooled perturbation screening, epigenetic editing methods (CRISPRa/i/on/off), molecular biology, nucleic acid delivery methods

Team Level Key Performance Indicators:

• Cost per reprogramming hypothesis for library generation and delivery
• Number of high-priority hypotheses tested per perturbation experiment
• Dynamic range of reprogramming factor expression (induced vs. non-induced) and variation across cells
• Efficiency of reprogramming factor delivery to target cell types
